Description
JAK3 (Phospho-Tyr785)Colorimetric Cell-Based ELISA Kit
The JAK3 Phospho-Tyr785 Colorimetric Cell-Based ELISA Kit is specially designed for the accurate measurement of JAK3 phosphorylation at tyrosine 785 in cell lysates. This kit offers high sensitivity and specificity, allowing for reliable and reproducible results in a wide range of research applications.JAK3 (Janus kinase 3) is a critical protein involved in signal transduction pathways, particularly in the regulation of immune responses and inflammation. Phosphorylation of JAK3 at tyrosine 785 is known to play a key role in its activation and downstream signaling cascades.
Dysregulation of JAK3 phosphorylation has been implicated in various diseases, including autoimmune disorders and certain cancers.By accurately measuring JAK3 phosphorylation at tyrosine 785, researchers can gain valuable insights into the molecular mechanisms underlying these diseases and potentially identify new therapeutic targets. The JAK3 Phospho-Tyr785 Colorimetric Cell-Based ELISA Kit is a valuable tool for studying JAK3 signaling pathways and developing targeted therapies for related conditions.

The JAK3 (Phospho-Tyr785) Colorimetric Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can detect JAK3 protein phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated JAK3 in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on JAK3 phosphorylation.
Qualitative determination of JAK3 (Phospho-Tyr785) concentration is achieved by an indirect ELISA format. In essence, JAK3 (Phospho-Tyr785) is captured by JAK3 (Phospho-Tyr785)-specific primary (1ø) antibodies while the HRP-conjugated secondary (2ø) antibodies bind the Fc region of the 1ø antibody. Through this binding, the HRP enzyme conjugated to the 2ø antibody can catalyze a colorimetric reaction upon substrate addition. Due to the qualitative nature of the Cell-Based ELISA, multiple normalization methods are needed:
1. | A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. |
2. | Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density. After staining, the results can be analysed by normalizing the absorbance values to cell amounts, by which the plating difference can be adjusted. |

UniProt Protein Function: | JAK3: a non-receptor tyrosine-kinase of the Jak family expressed predominantly in immune cells. Mediates signal transduction via the common gamma-chain of cytokine receptors, including IL-2, -4, -7, -9, and -15. Interacts with members of the STAT (signal transduction and activators of transcription) family. Interacts with STAM2 and SHB. Contains two protein kinase domains. Mutations that abrogate JAK3 function cause an autosomal SCID (severe combined immunodeficiency disease). Inhibitor: R017s for organ transplants. Three differentially spliced isoforms have been described. |

NCBI Summary: | The protein encoded by this gene is a member of the Janus kinase (JAK) family of tyrosine kinases involved in cytokine receptor-mediated intracellular signal transduction. It is predominantly expressed in immune cells and transduces a signal in response to its activation via tyrosine phosphorylation by interleukin receptors. Mutations in this gene are associated with autosomal SCID (severe combined immunodeficiency disease). [provided by RefSeq, Jul 2008] |

1. | Seed 200 µL of 20,000 adherent cells in culture medium in each well of a 96-well plate. The plates included in the kit are sterile and treated for cell culture. For suspension cells and loosely attached cells, coat the plates with 100 µL of 10 µg/ml Poly-L-Lysine (not included) to each well of a 96-well plate for 30 minutes at 37 °C prior to adding cells. |
