The Human CD274 (PD-L1) Monoclonal Antibody (PE Conjugated, 29E.2A3) from Assay Genie (catalog number AGEL1649) is a cutting-edge tool for researchers studying the programmed death-ligand 1 (PD-L1) pathway, a critical regulator of immune responses and a key target in immunotherapy. This monoclonal antibody, derived from mouse hybridoma technology, specifically targets the CD274 protein (PD-L1) in human samples and is conjugated with phycoerythrin (PE) for easy detection and analysis. Validated for multiple applications including flow cytometry and immunofluorescence, this antibody enables precise identification and quantification of PD-L1 expression on various cell types.PD-L1, a major immune checkpoint molecule, is known to play a crucial role in immune evasion, allowing cancer cells to evade immune surveillance and enabling tumor growth.
Targeting the PD-L1 pathway has shown promising results in cancer immunotherapy, making this antibody an invaluable tool for researchers investigating PD-L1 expression in cancer biology, immunology, and drug development.By providing a high-affinity and specific binding to PD-L1, the Human CD274 Monoclonal Antibody (PE Conjugated, 29E.2A3) offers researchers a reliable means to study the mechanisms of immune checkpoint blockade in cancer and other immunological contexts, ultimately contributing to the development of novel therapeutic strategies for combatting cancer and other immune-related disorders.
Each lot of this antibody is quality control tested by flow cytometric analysis. The amount of the reagent is suggested to be used 5 µL of antibody per test (million cells in 100 µL staining volume or per 100 µL of whole blood). Please check your vial before the experiment. Since applications vary, the appropriate dilutions must be determined for individual use.

Background:
CD274, also known as PD-L1 and B7-H1, is type I transmembrane glycoprotein that serves as a ligand for CD279 (PD-1). This interaction is believed to regulate the balance between the stimulatory and inhibitory signals needed for responses to microbes and maintenance of self-tolerance. CD274 is involved in the costimulation of T cell proliferation and IL-10 and IFN-γ production in an IL-2-dependent and CD279-independent manner. Conflicting data has shown that CD274 can inhibit T cell proliferation and cytokine production, and alternatively, enhance T cell activation. Other studies suggest that CD274 may signal bidirectionally, raising interesting implications for its expression in a wide variety of cell types, including T and B cells, antigen-presenting cells, and nonhematopoietic cells.
