Background: | CD16 (Fc??RIII) is a 50-80 kDa glycoprotein serving as a low affinity IgG receptor. Human Fc??RIII is expressed in two forms Fc??RIII-A and ??B. Fc??RIII-A is a transmembrane protein of monocytes, macrophages, NK cells and a subset of T cells. It is associated with Fc?µRI-?? subunit and is responsible for antibody-dependent NK cell cytotoxicity. Mast cell Fc??RIII-A is associated, moreover, with Fc?µRI-?? subunit. Besides IgG, Fc?µRI-A can be triggered also by oligomeric IgE. Fc??RIII-B is a GPI-linked monomeric receptor expressed on neutrophils and is involved in their activation and induction of a proadhesive phenotype. |

NCBI Summary: | This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other other antibody-dependent responses. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia.
