Description
ATP1A1 (Phospho-Ser23)Colorimetric Cell-Based ELISA Kit
The ATP1A1 Phospho-Ser23 Colorimetric Cell-Based ELISA Kit is a powerful tool for quantifying levels of phosphorylated ATP1A1 (sodium/potassium-transporting ATPase subunit alpha-1) in cell culture samples. This kit offers high sensitivity and specificity, allowing for accurate and reproducible results that are essential for studying cellular signaling pathways.Phosphorylation of ATP1A1 at Ser23 is known to regulate the activity of the sodium/potassium pump, which plays a critical role in maintaining cellular homeostasis.
Dysregulation of ATP1A1 phosphorylation has been implicated in various disease states, including hypertension and heart failure.By using the ATP1A1 Phospho-Ser23 Colorimetric Cell-Based ELISA Kit, researchers can gain valuable insights into the role of ATP1A1 phosphorylation in cellular function and disease pathogenesis. This kit is suitable for a wide range of research applications and is an indispensable tool for studying signal transduction pathways and identifying potential therapeutic targets.

The ATP1A1 (Phospho-Ser23) Colorimetric Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can detect ATP1A1 protein phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated ATP1A1 in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on ATP1A1 phosphorylation.
Qualitative determination of ATP1A1 (Phospho-Ser23) concentration is achieved by an indirect ELISA format. In essence, ATP1A1 (Phospho-Ser23) is captured by ATP1A1 (Phospho-Ser23)-specific primary antibodies while the HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Through this binding, the HRP enzyme conjugated to the secondary antibody can catalyze a colorimetric reaction upon substrate addition. Due to the qualitative nature of the Cell-Based ELISA, multiple normalization methods are needed:
1. | A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. |
2. | Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density. After staining, the results can be analysed by normalizing the absorbance values to cell amounts, by which the plating difference can be adjusted. |

UniProt Protein Function: | ATP1A1: the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of Na and K ions across the plasma membrane. This action creates the electrochemical gradient of Na and K, providing the energy for active transport of various nutrients. Two splice-variant isoforms have been described. |

NCBI Summary: | The protein encoded by this gene belongs to the family of P-type cation transport ATPases, and to the subfamily of Na+/K+ -ATPases. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The catalytic subunit of Na+/K+ -ATPase is encoded by multiple genes. This gene encodes an alpha 1 subunit. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2009] |
