Background: | This gene encodes a Jumonji- and AT-rich interaction domain (ARID)-domain-containing protein. The encoded protein is a DNA-binding protein that functions as a transcriptional repressor. This protein interacts with the Polycomb repressive complex 2 (PRC2) which plays an essential role in regulating gene expression during embryonic development. This protein facilitates the recruitment of the PRC2 complex to target genes. Alternate splicing results in multiple transcript variants. UniProt Protein Function: | JARID2: a Polycomb protein required for neural tube formation. Contains a Jumonji C domain but lacks histone demethylase activity. Essential for normal heart development and function. Required for embryonic viability. Down-regulation of Jarid2 impairs ES cell differentiation and Oct4 silencing. Associates with the Polycomb-Repressive Complex 2 (PRC2), stimulating PRC2 methyltransferase activity. PRC2 includes the histone H3 Lys 27 (H3K27) methyltransferase Ezh2, EED, SUZ12, RBBP4 and RBBP7 and possibly AEBP2. The recruitment of PRC2 to a promoter leads to increased levels of di- and trimethylation of H3K27 (H3K27me2/3), regulating the balance between self-renewal and differentiation of embryonic stem cells. The ability of Jarid 2 to be recruited to DNA in ES cells is interdependent on PRC2. Its knockdown reduces PRC2 at its target promoters, while ES cells lacking the PRC2 component EED are deficient in Jarid2 promoter activity. Acts as a transcriptional repressor of ANF by binding to both GATA4 and NKX2-5 and repressing their transcriptional activator activities. Participates in the negative regulation of cell proliferation signaling. Interacts with the N-terminal region of GATA4 and the C- terminal region of the NKX2-5 homeobox and represses their ability to activate transcription of ANF. |
