![]() | Western Blot. Positive WB detected in: Hela whole cell lysate, PC-3 whole cell lysate, Raji whole cell lysate, U87 whole cell lysate, HepG2 whole cell lysate, Mouse liver tissue. All lanes: MAP4K4 antibody at 1:1000. Secondary. Goat polyclonal to rabbit IgG at 1/50000 dilution. Predicted band size: 143, 139, 151, 134, 142, 146 kDa. Observed band size: 143 kDa. |
![]() | IHC image of PACO63679 diluted at 1:100 and staining in paraffin-embedded human liver tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. |

Background: | Serine/threonine kinase that may play a role in the response to environmental stress and cytokines such as TNF-alpha. Appears to act upstream of the JUN N-terminal pathway. Phosphorylates SMAD1 on Thr-322. |
Synonyms: | Mitogen-activated protein kinase kinase kinase kinase 4 (EC 2.7.11.1) (HPK/GCK-like kinase HGK) (MAPK/ERK kinase kinase kinase 4) (MEK kinase kinase 4) (MEKKK 4) (Nck-interacting kinase), MAP4K4, HGK KIAA0687 NIK |
UniProt Protein Function: | HGK: a Ser/Thr kinase of the STE20 subfamily. May play a role in the response to environmental stress and cytokines such as TNF-alpha. May interact with the SH3 domain of the adapter proteins Nck. Binds, via its CNH regulatory domain, to the N- terminal region of SPG3A. Upregulated in primary tumors and tumor cell lines. Required for anchorage-independent growth and ras-dependent focus formation in multiple cell lines. 5 isoforms of the human protein are produced by alternative splicing. |
UniProt Protein Details: | Protein type:Protein kinase, STE; Kinase, protein; EC 2.7.11.1; Protein kinase, Ser/Thr (non-receptor); STE group; STE20 family; MSN subfamily Chromosomal Location of Human Ortholog: 2q11.2-q12 Cellular Component: cytoplasm Molecular Function:protein serine/threonine kinase activity; protein binding; ATP binding; receptor signaling protein serine/threonine kinase activity Biological Process: microvillus biogenesis; regulation of catalytic activity; protein amino acid phosphorylation; regulation of JNK cascade |
NCBI Summary: | The protein encoded by this gene is a member of the serine/threonine protein kinase family. This kinase has been shown to specifically activate MAPK8/JNK. The activation of MAPK8 by this kinase is found to be inhibited by the dominant-negative mutants of MAP3K7/TAK1, MAP2K4/MKK4, and MAP2K7/MKK7, which suggests that this kinase may function through the MAP3K7-MAP2K4-MAP2K7 kinase cascade, and mediate the TNF-alpha signaling pathway. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008] |
