Background: | FAT1, FAT2, FAT3 and FAT4 are human homologs of Drosophila Fat, which is involved in tumor suppression and planar cell polarity (PCP). FAT4 undergo the first proteolytic cleavage by Furin and are predicted to undergo the second cleavage by γ secretase to release intracellular domain (ICD). FAT4 directly interacts with MPDZ/MUPP1 to recruit membrane associated guanylate kinase MPP5/PALS1. FAT4 is involved in the maintenance of PCP and inhibition of cell proliferation. |
Synonyms: | FAT tumor suppressor homolog 4 (Drosophila) |
UniProt Protein Function: | FAT4: May function in the regulation of planar cell polarity. Cadherins are cell-cell interaction molecules. 3 isoforms of the human protein are produced by alternative splicing. |
UniProt Protein Details: | Protein type:Membrane protein, integral; Cell adhesion Chromosomal Location of Human Ortholog: 4q28.1 Cellular Component: apical part of cell; integral to membrane; intracellular; plasma membrane Molecular Function:calcium ion binding; protein binding Biological Process: cerebral cortex development; gut development; heart morphogenesis; heterophilic cell adhesion; homophilic cell adhesion; inner ear receptor stereocilium organization and biogenesis; neurogenesis; plasma membrane organization and biogenesis; ureteric bud branching Disease: Hennekam Lymphangiectasia-lymphedema Syndrome 2; Van Maldergem Syndrome 2 |
NCBI Summary: | The protein encoded by this gene is a member of the protocadherin family. This gene may play a role in regulating planar cell polarity (PCP). Studies in mice suggest that loss of PCP signaling may cause cystic kidney disease, and mutations in this gene have been associated with Van Maldergem Syndrome 2. Alternatively spliced transcript variants have been noted for this gene. [provided by RefSeq, Mar 2014] |
